Lets say if a organisation have the resource to use Bitlocker or Truecrypt.

Which one should they use to get the best security possible and why??

I'm not sure this is an answerable question for a couple reasons. "Best" in security is a nebulous concept. The best choice depends largely on your situation and there aren't any details about your situation here. What is the balance needed between usability and limiting exposure? Particularly when comparing two similar products, it is going to depend very largely upon your specific needs and which product provides for them the best. – AJ Henderson Dec 5 '12 at 19:37

Neither: you will be more secure if you do not store any data at all. If this is not an acceptable solution, then you have additional requirements. State them. What hardware will you be using (TPM or no TPM)? What software (Truecrypt works on all major platforms, Bitlocker requires Windows)? What physical security do you require? Who will the users be? Who will do the administration? How will users be sharing data? Is this for user machines or servers? How do you plan to recover data if a motherboard fries? if a disgruntled user leaves? … – Gilles Dec 5 '12 at 21:14
